Imine & Enamine Formation
Connect the choice of amine to C=N formation or alpha-carbon unsaturation.
1. Understand the Concept
A primary amine adds to an aldehyde or ketone, and after proton transfers and dehydration an imine (C=N) can form. A secondary amine under suitable mildly acidic conditions commonly produces an enamine (C=C–NR₂) when an alpha hydrogen is available. Water removal helps shift the equilibrium.
Learning outcome: Predict imine formation from primary amines and enamine formation from secondary amines when an alpha hydrogen is available.
2. Follow a Consistent Strategy
- Identify whether the amine is primary (RNH₂) or secondary (R₂NH).
- Use mild acid catalysis to promote addition and dehydration while retaining enough unprotonated amine.
- For a primary amine, predict an imine with a C=N bond.
- For a secondary amine with an alpha H, predict an enamine with C=C adjacent to nitrogen.

4. Work Through Examples
Primary amine and ketone
CH₃COCH₃ + CH₃NH₂ ⇌[H⁺] (CH₃)₂C=NCH₃ + H₂O
A methyl-substituted imine forms by net loss of water.
Secondary amine and ketone
Cyclohexanone + pyrrolidine ⇌[mild H⁺] cyclohexenyl-pyrrolidine + H₂O
The product is an enamine because the amine has no N–H remaining to form a neutral C=N imine.
5. Avoid These Mistakes
- Assuming very strongly acidic conditions always give faster reaction; fully protonated amine loses nucleophilicity.
- Calling an enamine an imine: an enamine contains C=C–N.
- Ignoring alpha hydrogens and regioisomers in enamine formation from unsymmetrical ketones.
6. Review & Check Understanding
What type of amine typically forms an imine?
A primary amine (or ammonia, forming an unsubstituted imine).
What structural feature is necessary for enamine formation?
An alpha hydrogen on the carbonyl substrate, in addition to appropriate secondary amine conditions.
